**Key characteristics** * CD37 is a glycoprotein with a molecular weight of approximately 40-50 kDa. * It is expressed on the surface of professional antigen-presenting cells, such as B cells and macrophages. * CD37 is also expressed on activated T cells, natural killer (NK) cells, and activated dendritic cells. * It is a homophilic interaction between CD37 on a cell and CD37 on another cell. **Pathways and functions** * CD37 is involved in the regulation of B cell activation and differentiation. * It is expressed on B cells that are in the early stages of differentiation and are receiving signals from T cells. * CD37 signaling can inhibit B cell activation, differentiation, and cytokine production. * It is also expressed on mature B cells that are in the plasma membrane. * CD37 signaling can promote B cell activation and survival. **Clinical significance** CD37 is a promising target for cancer immunotherapy. Antibodies against CD37 have been shown to be effective in blocking B cell activation and inhibiting tumor growth in mouse models of cancer. Clinical trials of CD37 inhibitors are currently underway in humans with several types of cancer.
