**Key characteristics:** * EDN3 is a cell surface receptor for endothelin. * It is a member of the Class a/1 (rhodopsin-like receptors) of cell surface receptors. * It is expressed in high levels in blood vessel endothelial cells, enteric smooth muscle cells, and cardiac endothelial cells. * It is also expressed in low levels in epithelial cells of the lower respiratory tract and goblet cells. **Pathways and functions:** * EDN3 is a key regulator of blood pressure. It does this by binding to endothelin on the surface of blood vessels and promoting the formation of the endothelial cell adhesion molecule (ECAM) complex. The ECAM complex is a bridge between the endothelial cells and the smooth muscle cells that line the blood vessels. When the ECAM complex is formed, it allows the smooth muscle cells to contract and relax, which can affect blood pressure. * EDN3 also plays a role in maintaining blood vessel integrity. It does this by binding to endothelin and promoting the assembly of a complex called the endothelial cell adhesion molecule (ECAM) complex. The ECAM complex is a network of adhesion molecules that holds the endothelial cells together and prevents them from dislodging. * EDN3 also plays a role in regulating inflammation. It does this by binding to endothelin and promoting the migration of immune cells into the blood vessel. This migration can help to clear infection and inflammation, but it can also contribute to the development of atherosclerosis. **Clinical significance:** EDN3 is a promising target for the treatment of a variety of diseases. For example, EDN3 mutations have been linked to the development of hypertension, atherosclerosis, and heart failure. Targeting EDN3 with small molecules is a promising way to treat these diseases.
