**Key characteristics:** * KIR2DL4 is a type I transmembrane receptor that belongs to the killer cell immunoglobulin-like receptor (KIR) family. * It is composed of two Ig domains and a long cytoplasmic tail. * KIR2DL4 is expressed on the surface of hematopoietic stem cells, decidual natural killer cells, and some immune cells. * It is also expressed on a variety of epithelial cells, including enterocytes, goblet cells, and intestinal crypt stem cells. **Pathways and functions:** * KIR2DL4 is a negative regulator of natural killer cell mediated cytotoxicity. * It interacts with the immune checkpoint receptor PD-1, which is expressed on natural killer cells. * By binding to PD-1, KIR2DL4 inhibits the activation of the immune checkpoint receptor and allows natural killer cells to kill target cells. * KIR2DL4 also plays a role in the regulation of cellular senescence. **Clinical significance:** * Kir2DL4 mutations are associated with several human diseases, including cancer and autoimmune disorders. * Inhibition of KIR2DL4 has been shown to enhance the antitumor activity of immune checkpoint blockade therapies. * Therefore, KIR2DL4 is a promising target for cancer immunotherapy.
